چکیده

The aim of this work was to study the sarcomeric organization of muscles with PSE-like zones and normal muscle (control), focusing on the SHG signal pattern of myosin (single or double band). Second harmonic generation microscopy has been applied on pork Semimembranosus samples fixed at 24 hours post mortem. Electronic microscopy was applied on the same samples to improve SHG images interpretation. PSE-like zones were presenting SHG signal with a higher single band frequency than the control muscles (87.5% vs 35%, respectively). The defect SHG pattern was close to the data obtained on skeletal muscle in normal physiological conditions. This result indicates that the sarcomeric alignment is mostly maintained in PSE-like zone whereas the alignment is over in normal muscle. Electronic microscopy images confirmed this data. This study reveals that SHG microscopy could be a new tool to identify structure defects in pork muscles at 24h postmortem, in addition to the subjective PSE-like zone scale.

Impact of Chilling Rate and Halothane Genotype on the Frequency of Pse-like Zones and the Processing Yields of Hams

The aim of this work was to further investigate the effects of chilling rate on the frequency of PSE-like zones on pork hams, following on from a previous study [1]. An 8°c difference in chilling rate (4 hours post-mortem) was applied to the opposite carcass sides, from halothane genotyped pigs. In order to increase the frequency of PSE-like zones, pigs were slaughtered after a limited fasting ...

Mapping of the organization of p-nitroaniline in SAPO-5 by second-harmonic generation microscopy.

Second-harmonic generation microscopy (SHGM) has been employed to study crystals of zeolite-like material SAPO-5 filled with p-nitroaniline (PNA). The SHG and 2-photon fluorescence response of PNA in the one-dimensional channels readily reveals the pore accessibility; intergrown crystallites containing hexagonal pyramidal components and internal diffusion barriers are found next to seemingly pe...

Muscle metabolism and PSE pork

Pale, soft, and exudative (PSE) pork is primarily caused by an accelerated rate of postmortem glycolysis resulting in low muscle pH while carcass temperature remains high, thus causing protein denaturation. Numerous factors influence the rate of postmortem metabolism and may be responsible for the rapid pH decline characteristic of PSE pork during the 1st h postmortem. Release of high levels of...

Comparison of protein degradation, protein oxidation, and μ-calpain activation between pale, soft, and exudative and red, firm, and nonexudative pork during postmortem aging.

The objective of this study was to investigate the differences in protein modifications between pale, soft, and exudative (PSE) and red, firm, and nonexudative (RFN) pork during postmortem (PM) aging. Longissimus dorsi (LD) including 8 PSE and 8 RFN muscles were individually removed from 16 carcasses. These 16 LD muscles were vacuum packaged at 24 h after slaughter and stored at 4°C for 1, 3, a...

Stepwise chilling: tender pork without compromising water-holding capacity.

The current pork slaughter process is primarily optimized to reduce cooler shrink and the incidence of PSE pork. Elimination of the halothane gene and improved preslaughter handling have decreased the incidence of PSE pork and improved the water-holding capacity of the muscle; however, the chilling process has not been optimized to accommodate these changes. The hypothesis that stepwise chillin...
